I would like to take a survey of where you are in your process and what difficulties you have had. So I will post something on the Blackboard this weekend.
Our first DNA isolation went fairly well. Although there was a little bit of effort required to push Josh’s DNA and wash through the column,nothing like what we found in our trial run (pic). Have some rest and relaxation this weekend,thinking about the free energy that allows you do so. Yesterday we took a pic of each plate. We have 22 unique plaque morphologies and 2 more on the way. This is a pic of an isolation plate.  Today’s notebook checks brought several joys. Highlighting mistakes and underlining important steps was a nice touch. We should work on stating why we are doing what we are doing,clearly labeling the results,and including pictures. Another joy is seeing the clear variations of plaque morphology. We don’t know why there are no phage in the soils from Lubbock. Among the joys of today… Team 3 multitasking- blogging and plating at the same time!! A skeptic may say- but is learning taking place? I am usually a skeptic,but there is no doubt that learning is happening. How many instructors can say that after only a few weeks their students are invested in the outcome of a course? BEARS in the SEA are hungry. Do they need to be fed a little more meat? Are you ready to explore what is happening when that plaque appears? So,as we build our tapestry of what research looks like,one thread will definitely be MIXING. Mix the dilutions and mix the TOP AGAR- and keep it at 55. The joy was to watch as the phage hunters repeated their experiment with more confidence and skills! Now I am back to anticipation. Just to add to your suspense,I am not feeling real good about the top agar. We waited until after 5 and it was still slushy. What could make the 1X top agar runny? Think about that and we will talk about it tomorrow. Where did the week go? I had a professor once tell me that ‘time was fluid’. I think that he said that because he was always running late and had a hard time remembering meetings. Events just ran together for him. Hmm… Time was fluid for me this week. Many joys,but I particularly liked the good natured humor from our tardy,fun loving phage hunter that was denied both a Resource Manual and a Lab Notebook! My daughter is in her 7th month of pregnancy. This is our first grandchild. I am reliving the joys of anticipation and remembering the joys of seeing a baby for the first time and watching him or her grow and develop. It has been a long gestation and I can’t wait to see this “baby”and watch her grow! | |
